چکیده

Thank you again for submitting your manuscript for consideration by The EMBO Journal. It has now been seen by three expert reviewers, whose comments are copied below. As you will see, they all consider your results as a first implication of K11-linked ubiquitin chains in TNF signaling interesting and potentially important and thus in principle suited for publication in The EMBO Journal. Nevertheless, they also all raise a number of major as well as more specific issues with the manuscript in its current form. While I do not feel that all of them will necessarily have to be addressed through further experiments to warrant successful further consideration, one point raised especially by referees 2 and 3 will however be very important, which is to provide stronger and more direct evidence for the actual involvement/importance of K11-linked chains in TNF/NF-kB signaling. On the other hand, some of the more specific points (such as extending certain experiments also to homologs/paralogs) that would round up the analysis may not be strictly required, unless of course unraveling potential redundancies might help in uncovering the physiological significance of the TNF-induced K11 chains as mentioned above.

c-IAP1 and UbcH5 promote K11-linked polyubiquitination of RIP1 in TNF signalling.

Ubiquitin ligases are critical components of the ubiquitination process that determine substrate specificity and, in collaboration with E2 ubiquitin-conjugating enzymes, regulate the nature of polyubiquitin chains assembled on their substrates. Cellular inhibitor of apoptosis (c-IAP1 and c-IAP2) proteins are recruited to TNFR1-associated signalling complexes where they regulate receptor-stimula...

K11-linked polyubiquitination in cell cycle control revealed by a K11 linkage-specific antibody.

Polyubiquitination is a posttranslational modification where ubiquitin chains containing isopeptide bonds linking one of seven ubiquitin lysines with the C terminus of an adjoining ubiquitin are covalently attached to proteins. While functions of K48- and K63-linked polyubiquitin are understood, the role(s) of noncanonical K11-linked chains is less clear. A crystal structure of K11-linked diubi...

c-IAP1 binds and processes PCSK9 protein: linking the c-IAP1 in a TNF-α pathway to PCSK9-mediated LDLR degradation pathway.

Recent genetic studies have shown that PCSK9, one of the key genes in cholesterol metabolism, plays a critical role by controlling the level of low-density lipoprotein receptor. However, how PCSK9 mediates LDLR degradation is still unknown. By combining a shotgun proteomic method and differential analysis of natural occurring mutations of the PCSK9 gene, we found that an E3 ubiquitin ligase c-I...

OTUB1 modulates c-IAP1 stability to regulate signalling pathways.

The cellular inhibitor of apoptosis (c-IAP) proteins are E3 ubiquitin ligases that are critical regulators of tumour necrosis factor (TNF) receptor (TNFR)-mediated signalling. Through their E3 ligase activity c-IAP proteins promote ubiquitination of receptor-interaction protein 1 (RIP1), NF-κB-inducing kinase (NIK) and themselves, and regulate the assembly of TNFR signalling complexes. Conseque...

A20 ubiquitin ligase-mediated polyubiquitination of RIP1 inhibits caspase-8 cleavage and TRAIL-induced apoptosis in glioblastoma.

UNLABELLED The TNF-related apoptosis-inducing ligand (TRAIL) apoptotic pathway has emerged as a therapeutic target for the treatment of cancer. However, clinical trials have proven that the vast majority of human cancers are resistant to TRAIL apoptotic pathway-targeted therapies. We show that A20-mediated ubiquitination inhibits caspase-8 cleavage and TRAIL-induced apoptosis in glioblastoma th...
